Production and Clinical Significance of insulin antibody in Monosulin-treated Diabetic patients







Abstract
To evaluate the safety and the efficacy of Monosulin treatment in diabetic subjects, insulin antibody production and other side effects were studied in 63 noninsulin dependent diabetic patients who treated by Monosulin for 6 months.
The quantification of circulating insulin antibodies were performed by equilibrium binding assay with 1251 - monoiodinated insulin. The anti-insulin antibody was detected from 14.7% of total blood samples which were obtained during treatment with Monosulin. The positive rate of insulin binding was 11.1% at the first month of treatment and increased upto 22% after 6 month.
The mean percent bound insulin was 1.14% at the first month of treatment and increased to 1.64% at the second month. Thease mean percent bound insulin were quite lower than that of mean ¡¾2SD of normal population. The positive correlation between daily Monosulin (N.P.H) dosage and percent bound insulin was noted in this study and it may reflect preformed anti-insulin antibody interfere biologic action
of insulin.
The mean daily insulin requirement for satisfactory control of blood glucose was 28U by Monosulin N.P.H. There were only minor side effects such as a few case of drug eruption (were noted) during Monosulin treatment. Thease data suggested Monosulin treatment may be one of the useful modality for control of diabetes mellites.
